Cetrorelix Acetate (CAS 130143-01-0) is is a gonadotropin-releasing hormone (GnRH) antagonist that is marketed primarily under the brand name Cetrotide. A synthetic decapeptide, it is used is used in assisted reproduction to inhibit premature luteinizing hormone surges. The drug works by blocking the action of GnRH upon the pituitary, thus rapidly suppressing the production and action of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Cetrorelix Acetate is composed of a variety of non-natural amino acids that prevent early ovulation in clinical trials, leading to ovulation collection and assisted reproductive technology therapy for patients undergoing controlled ovarian stimulation. Indication Cetrorelix Acetate is an artificial hormone that blocks the effects of gonadotropin-releasing hormone (GnRH). In assisted reproductive technology, in patients with controlled ovarian stimulation, prevent premature ovulation. Pharmacological action Cetrorelix Acetate is a gonadotropin-releasing hormone (GnRH) antagonist. GnRH can bind to membrane receptors of pituitary cells. Cetrorelix acetate competitively binds to these receptors with endogenous GnRH, thereby dose-dependently inhibiting the secretion of luteinizing hormone (LH) and follicle stimulating hormone (FSH) by the adenohypophysis. The inhibitory effect was produced immediately after the administration of cetrorelix acetate, and the inhibitory effect could be maintained by continuous treatment. for women, Cetrorelix Acetate can delay the LH peak, thereby delaying ovulation. For women undergoing ovarian stimulation, the duration of action was dose-dependent. A single dose of cetrorelix 3mg was estimated to have an inhibitory effect of at least 4 days, with an inhibitory effect of approximately 70% on Day 4. Repeated administration of cetrorelix acetate 0.25mg every 24 hours can maintain its effect. The hormone-antagonizing effect of Cetrorelix Acetate was completely reversed at the end of treatment in both animals and humans. Biological activity Cetrorelix Acetate is a potent antagonist of the GnRH receptor with an IC50 value of 1.21 nM. Target IC50: 1.21 nM GnRH.
